Using an Enzyme to Amplify the Signal

The signal was amplified by attaching the enzyme alkaline phosphatase rather than a fluorescent marker to the antibody. Alkaline phosphatase catalyzes a chemical reaction, and one of the reaction products is a dark blue molecule that can readily be seen under the microscope.

Connected to the right of the probe is a blue shape labeled tag molecule (DIG) and a red Y shape next to it labeled Anti-DIG antibody. A pentagon labeled Alkaline phosphatase is connected to the anti-DIG antibody.
